Evrogen offers cDNA normalization kits designed to generate full-length-enriched double stranded (ds) cDNA with equalized concentrations of different transcripts. Both total RNA and poly(A)+ RNA can be used for cDNA preparation and subsequent normalization. Depending on your particular needs, you can select Trimmer or Trimmer-Direct kit wherein the first allows preparation of normalized cDNA ready for nondirectional cloning and the second – for directional cloning of cDNA library. Alternatively, DSN-normalized cDNA can be used for 454 sequencing. Important note regarding 454 sequencing.

Starting materials

Trimmer kit allows normalization of cDNA flanked by the same adapter sequences at the both 3'- and 5'- ends. Starting cDNA can be prepared using one of the following cDNA synthesis kits:

- Mint-2 cDNA synthesis kit (Evrogen Cat.# SK005)
- Mint cDNA synthesis kit (Evrogen Cat.# SK001)
- Mint-Universal cDNA synthesis kit, Protocol-I (Evrogen Cat.# SK002)
- SMART^TM PCR cDNA synthesis kit (Clontech Cat.# 634902).

Trimmer-Direct kit is designed for normalization of cDNA flanked by the different adapter sequences at the 3'- and 5'- ends. Starting cDNA must be prepared using one of the following cDNA synthesis kits:

- Mint-Universal cDNA synthesis kit, Protocol-II (Evrogen Cat.# SK002)
- SMART^TM cDNA Library Construction kit (Clontech Cat.# 634901)*
- Creator^TM SMART^TM cDNA Library Construction kit (Clontech Cat.# 634903)*.
